Występowanie przeciwciał dla rekombinowanych białek i lipopolisacharydów werotoksycznych pałeczek Escherichia coli (VTEC) w poszczególnych podklasach IgG u dzieci z zespołem hemolityczno-mocznicowym
[Serum immunoglobulin IgG subclass distribution of antibody responses to recombinant proteins and lipopolysaccharides of verocytotoxin-producing Escherichia coli (VTEC) in children with haemolytic-uraemic syndrome]

STRESZCZENIE

Określono częstość występowania i poziom przeciwciał podklas IgG1-IgG4 dla LPS oraz rekombinowanych białek werotoksycznych pałeczek E. coli u dzieci z zespołem hemolityczno-mocznicowym. Przeprowadzone badania wykazały, że w przebiegu zakażenia VTEC najczęściej wykrywano przeciwciała podklasy IgG1 i IgG2 dla LPS, następnie dla intyminy i białka Tir oraz dla werotoksyny 2b. Przeciwciała podklasy IgG3, a zwłaszcza podklasy IgG4 dla rekombinowanych białek w badanych próbkach surowicy diagnozowano znacznie rzadziej.

ABSTRACT

Introduction: The present study was aimed at determining the IgG subclass distribution against LPS and recombinant proteins of verocytotoxin-producing E.coli (VTEC) in children with haemolytic-uraemic syndrome.

Materials and Methods: The total number of 17 serum samples obtained from children with bacteriologically and/or serologically confirmed VTEC infections were tested by in-house ELISA for the presence of IgG1-IgG4 antibodies. As the antigen in ELISA were used LPS of E. coli serotype O157, O26, O111, O121, O145 and recombinants proteins: verocytotoxin 2b, intimin and Tir.

Results: In the present study most frequently and in the highest levels antibodies of subclass IgG1 and IgG2 to LPS, intimin and Tir protein were diagnosed. The arithmetic means of OD450 of antibodies IgG3 and IgG4 to these antigens were over two-times lower than antibodies IgG1 measured in all of tested serum samples. Antibodies of subclass IgG1, IgG2, IgG3 and IgG4 in diagnostically significant level to verocytotoxin 2b were diagnosed only in 8, 3, 4 and 3 serum samples for 17 tested, respectively.

Conclusions: In conclusion, IgG1 and IgG2 antibodies to LPS, intimin and Tir protein are predominating IgG subclass diagnosed in children with haemolytic-uraemic syndrome.
