Streszczenie

Szybka i wiarygodna identyfikacja B. anthracis jest kluczowa dla podjęcia skutecznej terapii osób narażonych na kontakt z przetrwalnikami tego drobnoustroju. Zastosowanie technik biologii molekularnej znacząco skraca czas uzyskania wyniku. Jednak molekularna identyfikacja jest utrudniona przez wysokie podobieństwo genetyczne laseczek z grupy B. cereus. Wiele opracowanych testów okazało się nieswoistych. Jednakże jak dotychczas swoistość opisanego w 2007 roku testu RSI-PCR (Restriction Site Insertion - PCR) dla genu plcR do identyfikacji laseczki wąglika nie została podważona. W niniejszej pracy przedstawiono możliwość znacznego skrócenia czasu potrzebnego na uzyskanie wyniku w tym teście poprzez zastosowanie nowoczesnych, „szybkich” polimeraz i enzymów restrykcyjnych. Dzięki użyciu tego typu odczynników czas uzyskania wyniku metodą RSI-PCR dla genu plcR wynosi około dwóch godzin.

Abstract

Fast and reliable identification of B. anthracis is crucial for a successful therapy of persons exposed to anthrax spores. Use of molecular biology techniques significantly reduces time necessary for obtaining results. However, the molecular identification is hampered by the high genetic similarity of the B. cereus group bacteria. A lot of published B. antharcis identification approaches turned out to be non-specific. Nevertheless, the plcR-targeted RSI-PCR assay described in 2007 is still regarded
as highly specific for anthrax identification. In this paper possibility of significant reduction of time necessary for obtaining results by the use of modern, “fast” polymerases and restriction enzymes will be presented. The use of a such reagents enable to reduce time of the plcR-targeted RSI-PCR assay to about two hours
