ABSTRACT

Introduction: Staphylococcus aureus is the most frequently recognized microorganism responsible for hospital infections.

Materials and Methods: Thirty five genomes were aligned and screened for the presence of variable regions suitable for PCR in order to design a new genotyping method. For comparative analysis, SmaI-REA-PFGE and MLVA were used.

Results: Twenty new regions with various lengths were identified from multiple sequence alignment of the genomic DNA. Subsequently, 20 couples of primers were designed for the amplification of variable regions for the PCR method. Comparative analysis of both PCR genotyping methods and SmaI-REA-PFGE revealed higher number of genotypes for new method with the use of 35 genomic sequences.

Conclusions: The simplicity and easy interpretation of the results in the designed PCR method using 20 new variable regions makes it an attractive alternative to SmaI-REAPFGE and MLVA.
